  • The number of rheumatoid arthritis females was more than males and most common after age of 40thand 50thyears.
  • The number of patients which living in urban areas was higher than those in rural areas.
  • Physical and chemical analysis of SF revealed more than 1 ml, yellow green color, cloudy, low viscosity, with large fat droplets, bloody fluid, low pH degree, low glucose level, high total protein and albumin levels
  • The reference value of latent-MMP-1 activity level was in the range of (47.38- 227.72) ng/ml.
  • Also, the activity level of enzyme was significantly different between the synovial fluid and serum of patients with RA and between the serum of patient’s RA and the serum of control.
  • Factors of age, gender and geographic place had no impact on the activity level of Latent MMP-1, while smoking factor had an impact on the activity level of enzyme in RA patients and control.
  • There were highly significant levels of sialic acid, MDA and significantly low levels of hyaluronidase activity, GSH, vitamin C in synovial fluid when compared with the serum of RA patients. Moreover, there was a highly significant increase levels of hyaluronidas, ALP, lactate dehydrogenase, sialic acid, MDA, sulfate, phosphorus, in contrast to a significant decrease of GSH, vitamin C, vitamin D3, calcium and calcium/phosphorus ratio in the serum of RA patients when compared with the serum of control.
  • A significant difference was found in levels activity of latent-MMP-1, CRP, hyaluronidase, ALP, sialic acid, GSH, MDA, Vitamin C, Vitamin D3, calcium, phosphorus and calcium/phosphorus ratio concentration between severe disease activity and moderate disease activity in RA.
  • A positive significant correlation between latent-MMP-1 and DAS-28 in serum and synovial fluid, while a negative significant correlation of latent-MMP-1 with vitamin C and Vitamin D3 in serum of RA patients was obtained.
  • Two porteinous components had been isolated by gel filtration chromatography.
  • It was found that only the first peak has high level activity of latent MMP-1.
  • The molecular weight of MMP-1 serum and synovial fluid using gel filtration chromatography was found to be (47.6 and 48.2) kilo dalton respectively, and by using SDS-electrophoresis was found to be (49.6 and 50) kilo dalton in serum and synovial fluid respectively.
  • The appearance of the main top of the enzyme from serum and SF at retention times (1.65) minutes compared with retention time of standard latent MMP-1 at (1.571) minutes.
  • Finally, the results revealed that the addition of ethylene diamine tetra acetic acid, 2,4-dinitrophenol, sodium azide, potassium oxalate, mercaptoethanol, glutathione, cystein and blood serum decreased the activity level of MMP-1,
  • while the addition of NaCl, MgSO4, CoCl2, CaCl2, CuSO4, ZnSO4, HgCl2and Pepsin increased the activity level of latent MMP-1.
