A doctoral thesis discussing a phenotypic and molecular study in the diagnosis and inhibition of enterotoxins of Staphylococcus aureus
On Wednesday, December 26, 2022, the College of Science at the University of Mosul discussed the doctoral thesis entitled “A phenotypic and molecular study in the diagnosis and inhibition of enterotoxins of Staphylococcus aureus isolated from different sources”, part of which was attended by Associate Professor Dr. Mohamed Sobhi and the administrator Dr. Omar Adel and a number of faculty members. The study presented by the student (Bushra Ali Kazem Hatem) in the Department of Biology / Microbiology Branch dealt with the investigation of the intestinal toxins of Staphylococcus aureus isolated from different sources and the study of the possibility of inhibiting these toxins at the phenotypic and molecular levels using a group of materials (plant extracts, chemical compounds and antibiotics). The study concluded that the largest proportion of these bacteria are producers of enterotoxins from various sources of isolation, and the initiators that were designed in this study succeeded in investigating the genes encoded for the production of enteric toxins and studying their genetic sequence.Five new genotypes of the Sea gene encoding enterotoxin type A were discovered in this bacterium, different from the genotype installed in the NCBI database. Mint was the most efficient in inhibiting these toxins, which opens new horizons in the possibility of using the substances that inhibit the intestinal toxins observed in the current study in therapeutic applications and food preservation.The study aimed to:Isolation of Staphylococcus aureus from different pathological and environmental sources and its diagnosis by traditional and modern methods. Investigate the ability of the isolated bacteria to produce enterotoxins by culture methods and their ability to produce enterotoxin type A by immunological methods as it is representative of the rest of the types of staphylococcal enterotoxins. Molecular investigation of the Sea gene encoding enteric toxin A in the studied isolates and study of the genetic sequence of this gene in isolates taken from different pathological and environmental sources. Selection of some substances to be used in an attempt to inhibit the production of enterotoxins in isolated bacteria and phenotypic investigation of the occurrence of inhibition by these substances.Studying the ability of selected inhibitors to inhibit the gene expression of the Sea gene and investigating the occurrence of gene expression inhibition using molecular methods.
