Master Thesis Viva-Biology Department
Master Thesis Viva in the College of Education for Pure Science entitled” Effect of using some Earthworms extracts on different types of pathogenic bacteria “The College of Education for Pure Science, University of Mosul, has done the Master Thesis Viva entitled ” Effect of using some Earthworms extracts on different types of pathogenic bacteria “,On Wednesday, November 23, 2022, the respected Dean of the College, Assistant Professor Dr. Qais Ismail Ibrahim, the Honorable Scientific Associate and Administrative Associate, the Honorable Head of the Department of Biology, and a number of the college’s teachers were attended the viva. This study, presented by the M.Sc. Student Hadeel Salem Ali Jassim Al-Shrefy in the Department of Biology, aims to find natural alternatives, especially animal ones, to inhibit the growth of types of Gram negative and Gram-positive bacteria For this, some extracts of the two sexes of earthworms Lumbricus terrestris and Aporrectodea tuberculate were used, The bioactive compounds were diagnosed in worms according to the GC-MS technique The results showed that the genus L.terrestris contains five chemical compounds that are effective in inhibiting the growth of bacteria Including the compound Thiodiglycol, which was estimated at (43.38%) and the compound Dimethoxyamine, which was estimated at (21.20%) while the second genus of earthworms is A. tuberculate, which contained 15 chemical compounds among these active compounds is the butan-(3-(1-Ethoxy-ethoxy) compound, which was found in the highest percentage (29.91%) it is toxic chemicals the compound 2-Aminoethyl-dl cysteine, which is an amino acid, is considered one of the mutant enzymes, with an estimated rate of 18.62%, In addition to the FTIR assay, which showed the presence of amide groups containing peptides in an undivided form, which led to the inhibitory activity against pathogenic bacterial isolates. The coelomic fluid of earthworms. L. terrestris and A. tuberculate showed inhibitory activity against pathogenic bacteria using the two solvents Petroleum Ether and Acetone, The coelomic fluid dissolved Petroleum Ether was more effective in inhibiting the growth of bacteria, and by comparison between the sexes, it was found that the coelomic fluid of the earthworm L.terrestris is more effective than the second sex.Extracts of earthworms were also made using alcoholic solvents (ethanol and methanol) and polar and non-polar solvents (acetone, hexane and chlorofoam), all of which had a significant and clear effect in inhibiting the growth of bacteria types used in the current study the results indicated that the best inhibition of bacterial growth was in the extract dissolved in acetone, and the highest inhibition rate was recorded in Salmonella bacteria, estimated at 29.33 mm at a concentration of 6.2 mg/ml, While in E.coli bacteria, the inhibitory activity was estimated at the same concentration of 24.67 mm, while the antibiotic recorded an inhibition rate that was less than the inhibition rate recorded by the acetone extract at the same sex bacteria, which was estimated at (22.33 and 0.0) mm, respectively, In comparison to the two worm extracts, it was observed that L. terrestris gave a higher inhibitory activity than A. tuberculate, as it contains mustard gas, which has a significant role in inhibiting the growth of most types of bacteria used in the current study. The results of the antibiotic susceptibility test showed that E.coli isolates were sensitive to Ciprofloxacin Tobramycin, and rate of 100% of the two antibiotics, while they were resistant to the antibiotics, Ampicillin, Azithromycin, Rifampin Tetracycline, and for K. pneumoniae bacteria they were rate of 100% sensitive to the antibiotics, Ampicillin, Azithromycin, Tobramycin Rifampin, Ceftriaxone, and Antibiotic Resistance, Chloramphenicol, Ciprofloxacin, Tetracycline. The inhibitory effect of all types of earthworm extracts was tested aforementioned against the types of bacteria used in the current study by method of drilling and method of testing turbidity and Lumbricus earthworm extract had a higher inhibitory effect than Aporrectodea earthworm extract, Also the effect of the extract increases by decreasing the concentration, especially at the concentration of 6.2 mg/ml.
