In fulfillment of the fourth goal of the Sustainable Development Goals (quality education) A doctoral dissertation entitled “A New Direct Molecular Identification of Streptomyces spp. Using Real-Time Polymerase Chain Reaction and Evaluating the Effect of Certain Culture Conditions on the Productivity of Their Metabolites” was discussed at the College of Education for Pure Science at the University of Mosul on Wednesday, June 18, 2025.
The study, presented by the student Intisar Hazem Hamed, from the Department of Biology, involved the extraction of 18 Streptomyces isolates, some of which were obtained from more than 60 samples from various locations, including buildings damaged by moisture, abandoned construction sites, and various soil types from various regions of Nineveh Governorate. These isolates were identified and purified using conventional laboratory methods, and the diagnosis was confirmed using polymerase chain reaction based on specific regions in srRNA16.
Sixteen soil samples were taken, and direct soil DNA isolation was performed based on specific Streptomyces genes within srRNA16. Conventional PCR and real-time PCR were used, using specific primers for each species. The conventional PCR results revealed the presence of one isolate among sixteen soil samples, while the real-time PCR results revealed the presence of twelve isolates among sixteen soil samples. This achieved the study’s aim of enabling diagnosis by direct DNA isolation using real-time PCR, without resorting to traditional isolation methods that are time-consuming and labor-intensive.
The discussion committee was chaired by Professor Dr. Hiyam Adel Ibrahim, and Assistant Professor Dr. Najdat Bahjat Mahdi, Assistant Professor Dr. Muhammad Abdullah Mahmoud, Assistant Professor Dr. Younis Saadi Saeed, and Assistant Professor Dr. Muhammad Arafat Muhammad as members, and Assistant Professor Dr. Jassim Fatehi Ali as both member and supervisor.
The Dean of the College presented a letter of thanks and appreciation to Assistant Professor Dr. Najdat Bahjat Mahdi from the College of Education for Pure Science, University of Kirkuk for his travel and participation in the student’s discussion.
