PH.D. Dissertation Viva in the College of Education for Pure Science entitled ” Inhibition of partially purified Kallikrein enzyme Activity from atherosclerosis patients with some Quercus fruit extracts and its effect on infected male rats “The College of Education for Pure Science, University of Mosul, has done the PH.D. Dissertation Viva entitled ” Inhibition of partially purified Kallikrein enzyme Activity from atherosclerosis patients with some Quercus fruit extracts and its effect on infected male rats “,On Wednesday, January 4, 2023, the respected Dean of the College, Assistant Professor Dr. Qais Ismail Ibrahim, the Honorable Scientific Associate and Administrative Associate, the Honorable Head of the Department of Biology, and a number of the college’s teachers were attended the viva. The current study, presented by the PH.D. Student Intisar Ghanim Taha Al-Sumaida’i in the Department of Chemistry, included four parts, first of clinical study of 165 people, 50 healthy (control) and 115 atherosclerosis patients (males and females) with ages ranging between 40-70 years. The enzymes estimated are PK, Casp-3, LOX, CAT, ALT and AST, also lipid profile T-Cho, TG, HDL-c, LDL-c, VLDL-c and atherogenic factor LDL-c/HDL-c were estimated. Other parameters BK, Nitric oxide, GSH, MDA, Glucose, urea, creatinine and finally Zn, Cu, Co, Pb, Mg and Ca were estimated.The PK, Casp-3, LOX, AST enzymes, T-Cho, T.G, LDL-c, VLDL-c, and atherogenic factor, Ca, ONOO, MDA, urea and creatinine found a significantly increased p≤0.05 in patients compared to control. Whereas found a significant decrease in the levels of Cat, HDL-c, Mg, and GSH at p≤0.05 in patients compared to control.The parameters ALT, Cu, Co, BK and Glu showed a non-significant increase, only Zn metal decreased non-significantly.The effect of sex was studied on this parameter and showed a significant increase in PK, T-Cho, LDL-c, and Pb, with a decrease in Mg levels in males compared to females, while LOX activity was increased in females. Also, patients divided into two groups aged 45-55 and 56-65 were found a significantly increased in PK, Casp-3, TG, Pb and creatinine with a decrease in CAT and Mg as getting older.The second part of this study included partially purified PK enzyme from the blood plasma of atherosclerosis patient by several techniques, including ammonium sulfate precipitate, dialysis, ion exchange chromatography and Gel filtration, one proteinous enzyme peak was obtained with specific activity 1.37, 5.82, 21.87 and 46.8U/mg protein respectively. One peak also was obtained by applying the electrophoresis technique and the molecular weight of approximately 102Kd, while by Gel filtration was 91.2Kd.Also, in this part, the natural compounds Oils, flavonoids and saponin from Quercus fruit were isolated and identified by CGC and HPLC techniques respectively and found a good content of polyphenol compounds and unsaturated fatty acids.The third part was foxed on the study of the effect of isolated Oils, flavonoids and saponin on PK activity at 2000, 6000 and 10000ppm respectively, the result showed non-competitive by Oil and Saponin but it was nor-competitive by polyphenol extracts.The fourth part deals study the effect of extracts on PK activity and some biochemical parameters in induced atherosclerosis (by 500mg/kg of cholesterol in coconut oil) in rats throw 15days and the infection was confirmed through the histological study of the blood vessels and the heart of the rats and in the blood lipid profile. The active dose of the crude Quercus extracts was 250mg/Kg body weight. Finally, the effect of crude extract, oil, polyphenol and saponin extracts were studied in normal and atherosclerosis rats at 250, 7, 37.5 and 10 mg/Kg body weight respectively. The results showed a significant decrease in PK, ALT, AST, Casp-3 activity and the lipid profile T-Cho, T.G, LDL-c, and VLDL-c, whereas a significant increase the CAT activity and HDL-c in atherosclerosis rats compared to healthy at p≤0.05 in blood plasma.Finally, the effect of extracts in the liver and kidney showed a significantly increased in GSH and a significant decrease in MDA after 15 days of treatment with Quercus extracts.The Viva committee was chaired by Prof. Dr. omar yunus mohammad/Mosul Unvirsity /College of Education Pure Science and the membership of Prof. Dr. Firas shawqi abdul razzaq /Tikirit Unvirsity /College of Science, Asst.
