Master Thesis Viva in the College of Education for Pure Science entitled ” The Physiological and Histological Role of Gum Arabic in The Blood, Liver and Kidneys of Male Rats Treated with Carboplatin “The College of Education for Pure Science, University of Mosul, has done the Master Thesis Viva entitled ” The Physiological and Histological Role of Gum Arabic in The Blood, Liver and Kidneys of Male Rats Treated with Carboplatin “,On Monday, August 29, 2022, the respected Dean of the College, Assistant Professor Dr. Qais Ismail Ibrahim, the Honorable Scientific Associate and Administrative Associate, the Honorable Head of the Department of Biology, and a number of the college’s teachers were attended the viva. In this study, presented by the M.Sc. Student Marwah abdulmelik ahmed Alshorbaji in the Department of Biology, The results showed the therapeutic effect of Gum Arabic with a concentration of 15% on Rattus rattus norvigicus at the age of (2-3) months and with a weight of (200-250) after being injected intraperitoneal (i.p) with carboplatin (50 mg / kg) which is used to treat cancer and has serious side effects on patients with cancer, and its harmful effect depends on the amount of dose given and the duration, as it causes hematological toxicity, nephrotoxicity and hepatotoxicity, in addition to its therapeutic role. In addition to its therapeutic role, medical examinations are conducted before giving doses to patients, which depend on the patient’s age and health condition.The rats of the current study were divided into 4 groups, and each group contained 6 rats. The first group was the control group injected into the peritoneum with distilled water once, while the second group was injected with a single dose of distilled water into the peritoneum and was given Gum Arabic at a concentration of 15% after one day of Injections for a period of 30 days. the third group was injected with single dose of carboplatin (50 mg/kg) i.p, while the fourth group was injected with a single dose of carboplatin (50 mg/kg) i.p and after two day of injections, they were given Gum Arabic 15% with drinking water for 30 days. The blood was collected and the animals were sacrificed after 30 days. Our study demonstrated the therapeutic effect of Gum Arabic on the blood, kidneys and liver of rats injected with carboplatin and study the physiological, histological and biochemical changes by studying the kidney functions (blood urea, uric acid, creatinine and erythropoietin enzyme) and liver functions represented by (allanine aminotransferase, amino-aspartate aminotransferase, alkaline phosphatase and gamma-glutamyl transferase), as well as red blood cell count, white blood cell count, blood platelet count, Packed cell volume and hemoglobin in male rats in all groups and compared to the control group, in addition to studying the level of oxidative stress and antioxidants in the liver and kidneys of rats as a result of injecting them with carboplatin and then treating them with Gum Arabic.The results proved that Gum Arabic has an antioxidant role and as a treatment to reduce the side effects resulting from carboplatin to a large extent, as the Gum Arabic caused a significant decrease at the probability level p<0.05 in the concentration of blood urea, uric acid and creatinine, as well as a decrease in the level of liver enzymes alanine aminotransferase and aspartate aminotransferase and phosphatase Alkaline and gamma-glutamyl transferase, in addition, Gum Arabic reduced the level of malondehyde and increased the level of catalase and glutathione in the kidneys and liver of rats treated with carboplatin when compared to the control group. As well as an increase in the percentage of hemoglobin, red blood cells, white blood cells, platelets, the Packed cell volume and erythropoietin, as the results were very similar to the control group.The study showed that Gum Arabic reduced the risk of kidney injury and kidney failure and reduced renal tubular cell necrosis resulting from carboplatin, which led to a reduction in the level of blood urea, uric acid and creatinine in the blood serum, thus reducing histological abnormalities in the kidney tissue.Our study revealed an increase in the percentage of liver enzymes GPT, GOT, ALP, and GGT in the serum of rats treated with carboplatin, which is one of the main indicators of liver tissue damage as a result of cell necrosis and apoptosis and the leakage of their contents of enzymes into the bloodstream, thus raising the level of enzymes in the blood serum and increased the level of malondehyde as a result of lipid oxidation in the cell membrane and increased Kupffer cells inside the tissue as a result of tissue damage. Gum Arabic reduced the accumulation of carboplatin and reduced the level of malondehyde and Kupffer cells and maintained the level of oxidative enzymes because it is an antioxidant and anti-inflammatory, thus reducing the harmful effects of carboplatin on liver cells.
