Master thesis discusses the inhibitory effect of natural compounds in the expression of expeller pump genes in Pseudomonas aeruginosa bacteria

The Department of life sciences at the Faculty of Sciences of the University of Mosul discussed the master’s thesis tagged “the Inhibitory Effect of Natural Compounds on the Expression of Efflux Pump Genes in Pseudomonas aeruginosa”, submitted by researcher Zeina Salim Muhammad Najib, on Thursday, September 3, 2026. The aim of the study was to detect some genes of expelling pumps in Pseudomonas aeruginosa bacteria isolated from clinical sources in Nineveh governorate, to study their relationship with antibiotic resistance, as well as to evaluate the inhibitory effect of some natural compounds, in particular Thymol and cinnamaldehyde, in the expression of expelling pump genes. The study was conducted on 150 clinical samples collected from a number of hospitals in Mosul during the period from October 1, 2025 to January 20, 2026. The results showed bacterial growth in 80 samples by 53.3%, while no growth was shown in 70 samples by 46.7%. Among the samples with bacterial growth, 21 isolates of Pseudomonas aeruginosa bacteria were diagnosed, distributed to various clinical sources, including burn samples, ear swabs and urine samples. The results of Molecular Detection showed the presence of a number of ejection pump genes in the studied isolates, including mexB, mexd, Mexy, mexf, poprn, Emre, mfs2, PMPM, MLA and space, which belong to different families of ejection pumps, especially the RND system, which is one of the most important ejection pump systems associated with antibiotic resistance in P bacteria. aeruginosa. The results of the antibiotic sensitivity test also showed resistance to a number of antibiotics used in the study, reflecting the multiplicity of resistance mechanisms in P bacteria. aeruginosa. A representative isolate was selected for the testing of natural compounds, based on its possession of targeted expeller pump genes and the appearance of a high pattern of antibiotic resistance. The results of evaluating the effect of the two natural compounds Thymol and cinnamaldehyde showed an inhibitory effect when used in conjunction with some antibiotics, as the effectiveness of Meropenem against the selected isolation improved, as the inhibition area reached 23 mm when using thymol and 24 mm when using cinnamaldehyde, compared to meropenem used alone, to which the isolation showed resistance. The study also examined the effect of natural compounds on the expression of ejection pump genes, in order to assess the extent to which they can contribute to reducing one of the most important resistance mechanisms in P bacteria. aeruginosa. The results indicated the importance of studying natural compounds as potential adjuvants to reduce antibiotic resistance and enhance their effectiveness against resistant bacterial isolates.
