Doctoral dissertation discusses the molecular and biochemical evaluation of the exact deletion of the Y chromosome in infertile men

The Department of life sciences at the Faculty of Sciences of the University of Mosul discussed the doctoral thesis tagged “Molecular and biochemical evaluation of Y chromosome microdeletion in infertile male patients”, submitted by researcher Hanin Ethar Hashim, on Thursday, September 10, 2026. The study aimed to assess the levels of a number of hormones associated with male fertility, in addition to conducting a molecular study to evaluate six point mutations in three genes, namely GNAO1, AGTPBP1 and SPATA5, associated with sperm efficiency, and to study the effect of these mutations in the gene expression levels of these genes. The results showed the presence of statistically significant changes in the levels of a number of hormones in infertile men compared to the control group. At the molecular level, the results revealed the presence of point mutations in the AGTPBP1 and SPATA5 genes, indicating their association with disorders of spermatogenesis and efficiency. The study explained that the effect of the agtpbp1 gene may extend to the number of sperm, and is not limited to whip abnormalities, as the mutation in this gene has been associated with a stop in proliferative cells during the process of spermatogenesis, which may lead to a lack of sperm, as well as its role in sperm motility. The results also indicated that the structural defect resulting from the mutation in AGTPBP1 may be associated with a disturbance of signal transduction pathways via the GNAO1 gene, which saw a marked decrease in the level of gene expression, while the SPATA5 gene recorded a sharp increase in its gene expression, which may represent a compensatory mechanism for cell protection. The study concluded that there is a correlation between the molecular and hormonal axis associated with infertility in men, especially with Testosterone and prolactin hormone levels, which may contribute to building more integrated molecular and hormonal indicators to understand the causes and diagnosis of infertility.
